Description
Tryptophan Broth is a biochemical medium for detecting indole production and confirming presumptive Salmonella colonies.
Tryptophan serves as the substrate for tryptophanase-positive organisms. After incubation, Kovac's reagent produces a red-violet ring when indole is present; Salmonella confirmation cultures are expected to be indole negative.
A pure culture is inoculated and incubated at 36 ± 1°C for approximately 24 hours. Kovac's reagent is added and the surface colour reaction is interpreted according to ISO 6579.
