Why Clostridium perfringens testing is demanding
C. perfringens is anaerobic and spore-forming, a combination that makes bulk-cooked meat its ideal vehicle. Cooking drives off oxygen and kills competing flora while leaving spores viable; if the product then cools slowly through the growth range, the surviving spores germinate into a population with no competition.
The illness is toxin-mediated and occurs when large numbers of vegetative cells are ingested and sporulate in the gut, which is why the specification is a count against a threshold rather than absence. Small numbers are unremarkable; large numbers indicate a cooling failure.
Detection requires genuinely anaerobic incubation on tryptose sulfite cycloserine agar, where the organism produces characteristic black colonies. Inadequate anaerobiosis is the most common cause of a falsely low count — the organism will simply not grow.
