Description
Urea Broth is a liquid biochemical medium for differentiating microorganisms, especially Enterobacteriaceae, by urease production.
Urease hydrolyses urea and releases ammonia. The highly buffered formulation changes phenol red to pink-red when strong urease producers such as Proteus, Morganella and Providencia generate sufficient alkalinity.
A pure culture is heavily inoculated and incubated at 35 ± 2°C for eight to 48 hours. Turbidity with a pink-red colour change indicates a positive reaction.
