Description
Buffered Yeast Extract Broth is a non-selective liquid medium for enriching Legionella species from environmental samples.
Yeast extract supplies nutrients, alpha-ketoglutarate stimulates growth, ACES buffers the medium, and L-cysteine with ferric pyrophosphate satisfies the stated nutritional requirements of Legionella.
The broth is inoculated and incubated at 36 ± 2°C for up to ten days. Turbidity indicates growth, and the PDF describes confirmation by subculture to BCYE agar with and without L-cysteine. It notes that related BCYE agar is recommended by ISO 11731, but it does not assign a formal method reference to this broth.
