Description
Triple Sugar Iron Agar is a differential medium for identifying Enterobacteriaceae through carbohydrate fermentation, gas production and hydrogen-sulfide production.
Glucose, lactose and sucrose fermentation produces yellow acid reactions and may split the agar through gas production. Thiosulfate reduction reacts with ferric ions and produces a black precipitate.
A pure colony is streaked over the slant and stabbed into the butt, then incubated aerobically at 37°C for 24 hours. Slant, butt, gas and hydrogen-sulfide reactions are interpreted together.
